I'm assuming the Advanced Sentinel, the other character, and the Barrier are all on the same elevation (all grounded or all elevated), if so then the answer is yes.

From the Advanced Sentinel Rules:
Blocking terrain blocks a line of fire drawn to an Advanced Sentinel normally.

The reason I ask is because Sentinels are considered to be "elevated" (characters who are elevated can draw line of fire to them); and an elevated character can see over blocking terrain or barriers.

No, they are not elevated.

Colossal Size
(NON-OPTIONAL) Only characters with the symbol block lines of fire to or from this character. A character with this ability ignores the effects of hindering, elevated, and outdoor blocking terrain on movement. This character can’t be knocked back and can make ranged combat attacks against non-adjacent opposing character when they are adjacent to opposing characters. This character and a character on elevated terrain draw line of fire and make ranged combat attacks against each other as if they were both elevated. This ability can’t be countered.

As you can see, that only matters if the other character drawing LOF is already elevated.

Further more, even if the Sentinel was "elevated" (and it isn't), the blocking terrain would still block LOF between the "elevated" Sentinel and a normal, grounded character. From the BN rules, page 13:

LINE OF FIRE: THE RIM
A line of fire between an elevated character and a grounded character is blocked by grounded blocking terrain, elevated terrain (not occupied by...


Barrier will block LOF between a Sentinel and any grounded character unless that character has specific rules that say otherwise (like Susan Richards).

Two things, first the Sentinel is only treated as elevated when the other character is elevated.

From the Advanced Sentinel Rules:
An Advanced Sentinel and other characters on elevated terrain can make ranged combat attacks against each other as if they were on the same terrain level.

Second, even if the Sentinel was treated as elevated a grounded barrier between an elevated character and grounded character still blocks LOF.

From p. 13 of the Blackest Night Rulebook:
A line of fire between an elevated character and a grounded character is blocked by grounded blocking terrain....
