In a recent game I was using black costume spiderman to try out his defend power. Niether me nor the other guy I play with are really familiar with this power. I had spiderman next to sand man becasue sand man only has a fifteen I thought it would help. My friend then attacked and successfully hit sand man for three clicks of damage, so clicked him once. My friend then siad that since I used the "defend" power, I lose the invulnerability power. Is this true?

Not true at all. You still get your invulnerability as well as any other power you have. Defend works with everything.

No, that's not true. Defend does not replace other defensive powers, it just shares a Defense Value. Invulnerability will still work.

Also, you can "stack" Defend with Energy Shield/Deflection for example, so if you had Spider-Man with Captain America for example, Cap's defense would be 20 vs. Ranged Attacks.

The best way to look at Defend is to consider that person as a Defender. Any friendly character who is adjacent to a character with defend can share the defender's defensive value.

Defend will only change the defensive value. Sandman would keep Invulnerability and would share Spiderman's tasty 18 defence. It is a pain if you're trying to heal someone next to invisible girl. In short, you got ripped.

OUCH!, I hadn't thought of that. Just checked, though, and fortunately defend is optional. Turn it off when you go to heal, and it comes back on at the end of your turn, in time to defend (no pun intended) against any attacks your opponent launches.
