when a character uses a ladder to go to the roof, do they have to stop on the ladder? (hindering terrain effect on movement, or something)

No you don't have to stop on the ladder. It is considered clear terrain (like the stairs).
It does take 2 movement to change elevation using the ladders though.

The effect on movement would be to climb up the ladder. That would take 1 of the steps from the speed value for the change in elevation. See Stairs and ladders are considered clear terrain that allow characters to change their elevation level. and Climbing up or down ladders adds 1 square to the character’s movement cost. under Clear Terrain in the rulebook
